Abstract

The Trichinosis is a parasitary disease caused by the nematode Trichinella spiralis. During its implant in the host, it joins to intestinal wall, copulate and the larves newly born cross the epithelium arrive to blood torrent and lodge in the muscles. In continuous way, T. spiralis secrete a proteases and/or bioactives compound, which induces stress in the host cells, which can be measure trough the overexpression of a group of proteins, known as Heat shock proteins (Hsp), which are normal constituents of the cells and their synthesis is increased by the exposition to many forms of stress. The objective: of the present study was determinate if T. spiralis secrete bioactives compound in vitro, which action in MDCK epithelial cells induces stress, measure trough the expression of Hsp70. Methodology: Infect Long Evans rats with T. spiralis, sacrifice the rats and obtain T. spiralis by artificial digestion. The nematodes obtained are placed in culture medium and submitted to different temperature (4, 21 and 37°C), collecting the culture medium every 24 hours, which later is utilized on culture of MDCK epithelial cells. The analysis of Hsp70 expression in the MDCK cells was made by PAGE-SDS and Western blot techniques, and inmunodetected with anti-Hsp70 antibodies. Results: T. spiralis secretes bioactives compound, some of which induce that the host produces antibodies against them. The presence of these bioactives compound induces stress in the MDCK cells, which is measure trough the expression of Hsp70, being more evident its expression when is utilized the culture medium where was maintained the nematode to 37°C. At the same manner, was observed an increased of Hsp70 in the insoluble fraction, indicated us alterations in the cytoskeleton and its possible correction trough Hsp70. Archivos de Medicina
