Abstract
tween-20 is used during cell disruption to obtain Hepatitis B virus surface antigen (HBsAg). The specifications for HBsAg indicate that the quantity of Tween-20 allowed must not be more than 50 mg for each 100 mg of purified protein. Therefore, Tween-20 quantification must be determined before the release of each batch. This study proposes a spectrophotometric method to estimate the Tween-20 concentration in purified AgsHB samples, followed to the formation of a cobalt - polyoxyethylene complex, and its measure at 620 nm of optical density. This work focuses on the validation of this method.
