Changes in vascular and myometrial angiotensin II receptors in preeclampsia

Resumen

Increased sensitivity to the pressor effects of angiotensin II is a main feature of preeclampsia, although underlying mechanisms for this are poorly understood. AT2 receptors (AT2R) antagonize angiotensin II effects at AT1 receptors (AT1R) through a variety of mechanisms including receptor dimerization (AT1R/AT2R). In order to evaluate whether preeclampsia is associated with changes in the relative expression of angiotensin receptors and or changes in formation of the receptor dimer, we measured AT1R, AT2R and AT 1R/AT2R protein expression in samples of omental blood vessels and myometrium obtained from healthy (39.9 ± 0.37 weeks of pregnancy) and preeclamptic (36.1 ± 0.51 weeks of pregnancy) women (n=9) subjected to caesarean section. AT2R and AT1R/AT 2R dimer expression were found to be decreased in vascular tissue from preeclamptic patients. In contrast AT2R and the AT 1R/AT2R heterodimer were found to be increased in the myometrium of preeclamptic patients. These results suggest that vascular events in preeclampsia may be related to a decreased formation of AT 1R/AT2R heterodimer, while myometrial and vascular AT 2R seems to have different roles during pregnancy.
