Abstract

We studied activation of Pr1A, Pr1B, Pr1C, PR1D, PR1 E, Pr1F, Pr1G, Pr1H, Pr1I, Pr1J, and Pr1K isoforms from pr1 gen from Metarhizium anisopliae (Metschnikoff) Sorokin (Hypocreales: Clavicipitaceae) during the pathogenic process in Asian citrus psyllid, Diaphorina citri Kuwayama (Hemiptera: Psyllidae), adults. Ma57 and Ma62 strains were chosen because of their higher and lower pathogencity toward D. citri, respectively. Adult individuals were infected with a conidial suspension and fed on Murraya paniculata (L.) Jack (Sapindales: Rutaceae) branches during 7 days. Isoform activation during the pathogenic process were detected by RT-PCR at 10 different times after infection. Results indicated that Pr1A, Pr1B, Pr1C, Pr1E, and Pr1I isoforms were not activated at any time in either strain, while Pr1F and Pr1G isoforms were activated in both strains from 0 hour until 168 hours. Pr1D, Pr1H, Pr1J, and Pr1K isoforms were activated in different patterns and some of them intermittently. Activation pattern differences were observed in Pr1D, Pr1J, and Pr1K isoforms from Ma57 strain compared to Ma62 strain. The remaining eight isoforms presented a similar activation pattern in both strains. These results indicate that some isoforms are probably activated due to the infection process, other isoforms are activated independently from the infection process, and others play other roles in different processes. Our results contribute to the knowledge of the participation of the pr1 gene of M. anisopliae during the pathogenic process in the Asian citrus psyllid. However, more studies are needed to define the main role of pr1 gene isoforms in the pathogenicity of this fungus and its activity in pest control.
